This refers to the new covenant, of which Paul is a minister. Paul contrasts the old and new covenants. The new covenant is not a system of rules and regulations. Here "Spirit" refers to the Holy Spirit. It may also refer to the new covenant being "spiritual" in nature. Alternate translation: "the ministry that gives life because it is based on the Spirit"
